What is MOTS-C?

MOTS-C was first described in 2015 by Changhan Lee and colleagues at USC, who identified it as a peptide encoded within the mitochondrial genome's 12S rRNA region rather than nuclear DNA — one of a small class of "mitochondrial-derived peptides" discovered over the last two decades.

Proposed mechanism

In the original and subsequent rodent studies, MOTS-C translocates to the nucleus under metabolic-stress conditions and has been reported to activate AMPK signalling, a central regulator of cellular energy balance. This is the basis for its research framing as an "exercise-mimetic" peptide — mouse studies have looked at its effect on glucose uptake and insulin sensitivity under diet-induced metabolic stress.
